Here's the general process I use:
1. Write a good story as a script. Kepe in mind that it's for audio, so you want to make sure there's enough details in the dialogue or "god-perspective narration" to make sure it all makes sense.
2. Set up auditions. You want to have people who can do parts well, but also that can record nice, clean files.
3. Cast parts and have the people involved submit MP3 or WAV files of their lines.
4. Use a program like WaveLab to put all the lines in the right order.
5. Balance lines in terms of volume and left/right orientation.
6. Insert sound effects and balance accordingly.
7. Add music and set up fades in and out, plus volume accordingly.
8. Export as a final stereo MP3.'
That's about the size of it. I've got a tutorial on WaveLab editing up on Fanworks in the Features section. Really, the entire thing is just a matter of patience and persistence. It's far easier than making a fan film, and, I think, far more gratifying than simply writing a fan fiction text story.
